Iodinated contrast induced acute kidney injury in diabetic and nephropathy diabetic in rats

Abstract

The contrast-induced nephropathy (CIN) is directly linked to chronic diseases that compromise the oxygenation in the region of the renal medulla. Chronic hyperglycemia changes a hemodynamic in the microvasculature, endothelial dysfunction and activation of inflammatory processes and oxidative injury, incurring decreased in glomerular filtration rate (GFR). The aim of this study is to investigate the NIC at high risk factor such as diabetes mellitus (DM) and preexisting chronic renal disease and elucidate the role of the endothelial injury and renal microvasculature in this injury model. Material and Methods: Adult male Wistar rats and tubular cells, HK-2 e IRPTC, will be used. DM will be induced by streptozotocin and 5/6 renal ablation model (Nx) wil be performed. Cells will be cultured in hyperglycemic media. Renal function by inulin clearance and microalbuminuria, oxidative stress, expression and quantification of inducible nitric oxide synthase (iNOS), HO-1, HO-2, histology of renal tissue, analysis of genic expression of HO-1, HO-2 and synthase NO, quantification of NO, HK-2 and IRPTC cells transfection with siRNA to HO-1 and with plasmid for HO-1 will be performed.
