Anti-diabetogenic, antioxidant / antiinflammatory / antiapoptotic effects of aqueous extract of Passiflora alata and components vitexin, Isoorientin, rutin and catechin

Abstract

The search for new drugs from medicinal plants has motivated the research for alternative substances with potential anti-inflammatory that could aid in the treatment of inflammatory diseases, as in the case of type 1 diabetes. In the previous study, (International Immunopharmacology - title: Antioxidant and anti-diabetic potential of Passiflora alata Curtis aqueous leaves extract in type 1 diabetes mellitus (NOD-mice, in press, fapesp: 2011/06559-4) we have found that substances in the aqueous extract of leaves of Passiflora alata Curtis was able to decrease the incidence of the disease modulating the inflammatory mechanisms by decreasing the cellular infiltrate, apoptosis, oxidative stress, cellular infiltration, in the pancreatic islets. Moreover, we found the preserved gluthatione (liver and kidney) and insulin in this animal model. Additional properties, the decrease of the cell cycle regulatory proteins expression, such as CDK6, and increasing insulin levels in the pancreatic islets. CDK6 expression in infiltrative cells in the pancreatic islets and its reduction in the treated group suggest that flavonoids found in the leaves of P. alata have an anti-proliferative action and may have contributed to the inhibitory protein expression and subsequent cell cycle arrest of activated inflammatory cells (submitted). Thus, this study aims to evaluate the anti-inflammatory effects and the mechanisms by which the aqueous extract and the components vitexin, isorientin, catechin and rutin in vitro may have a regulatory property in cell cycle regulation, cell proliferation, oxidative stress and apoptosis in pancreatic islets submitted to mixed cell culture with activated T cell, as well as investing in the development of new drugs with the potential to interfere with the anti-inflammatory process and development of type1 diabetes.
