Role of adaptative immunity on the progress of ischemic heart disease after acute myocardial infarction

Abstract

The role of subtypes of lymphocytes in the progression of coronary heart disease may be a new strategic target in the therapy of myocardial infarction. The muscle loss following the coronary occlusion adds prognostic information and early reperfusion constitutes the key strategy for these patients. However, surrounding the necrotic core, an ischemic area is formed, that progresses to extension of the infarction or recovers, mainly during the first weeks after the ischemic insult. The determinants of cell viability are unknown, postulating that they arise from factors not only related to microcirculation or energy expenditure, but also to inflammatory and immune responses. Furthermore, the intense mobilization of progenitor cells secondary to myocardial infarction triggers large lymphocyte proliferation that colonizes plaques in development, contributing to recurrent ischemic outcomes. This project aims to better understanding of the immune and metabolic mechanisms involved. Specifically, we will study the adaptive immunity, with emphasis on lymphocytes subtypes involved in the early and late course of these subjects, including lipidomics and metabolomics, as well as the corresponding signaling pathways. Therapeutic pharmacological strategies and changes on intestine microflora will be evaluated up to 12 months. Finally, experimental study was designed to better understanding of the role of adaptive immunity on this scenario. (AU)
