Effects of Bifidobacterium animalis subsp. lactis HN019 and Bdellovibrios bacteriovorus HD100 on the experimental periodontitis in rats: microbiological, immunological, histological and genetic features of the host response

Abstract

The purpose of this study is to evaluate the effects of the probiotics Bifidobacterium animalis subsp. lactis HN019 and Bdellovibrios bacteriovorus HD100 on ligature-induce periodontitis in rats. 96 rats will be assigned to one of two groups: C (control) and PD (periodontitis). Each group will be divided into 6 subgroups according in the probiotic therapy (Subgroup HN019 - Bifidobacterium animalis subsp. Lactis; Subgroup HD100 - Bdellovibrios bacteriovorus) and administration route (Topic and Oral). PD will be induced with the placement of ligatures around first molars of each animal. The ligatures will be left in the same position during 14 days. In oral administration, the probiotics will be added to the drinking water of the animals for 28 days. In topic administration, the animals will receive 3 local applications of probiotics. All animals will be euthanized 28 days after the start of the study. The following parameters will be analyzed: i) bacterial microbiota (checkerboard DNA-DNA hybridization and qRT-PCR; ii) expression of pro- and anti-inflammatory cytokines, antimicrobial peptides, transmembrane proteins, cell surface receptor and transcription factor with immunoassays (ELISA and LuminexTM xMAP®), immunohistochemical reactions (streptavidin-biotin-peroxidase) and analysis of gene expression (qRT-PCR); iii) levels of alveolar bone loss and periodontal attachment loss with histomorphometric analysis; and iv) bone area, bone mineral density and bone volume (micro-CT). The data obtained will be subjected to statistical analysis (p < 0.05). (AU)
