Effect of tissue sealant based on snake venom and cyanoacrylates applied on acrylic resin's surface on quantity and activity of C. albicans cells, cytotoxicity on gingival fibroblasts (phase I) and surfaces degradation assay (phase II)

Abstract

Denture stomatitis is a common disease that affects denture wearers and its main cause is C. albicans. This microorganism easily adheres to acrylic resin, especially because its surface features, as high roughness and hydrophobicity. Initially, the aim of this study is to verify if four products (tissue sealant derived from snake venom, ethyl-cyanoacrylate, butyl-cyanoacrylate ou octyl-cyanoacrylate) are able to diminish C. albicans biofilm formation when used as coatings acrylic resin surface, assessed by microbiologic culture and XTT. Subsequently, to observe the influence of these products on C. albicans biofilm, cellular activity will be assessed by ROS assay. Furthermore, the products' cytotoxicity on human gingival fibroblasts by MTT assay will be evaluated due to the regular and direct mucosal-denture contact. Finally, to elucidate the mechanisms by which the products will reduce or increase biofilm's formation, coatings will be assessed for their physical integrity before and after chemical disinfection (1% sodium hypochlorite and 4% chlorhexidine gluconate) and biofilm growth on specimen for 14 and 30 days. This physical assessment will be done by surface's hydrophobicity (goniometer), mean surface roughness (surface confocal microscope- SCM) and surface's three dimensional images (SCM). The data will be analyzed differently for each test. Numerical data will undergo Shapiro-Wilk test and depending on the outcome will be examined by the pertinent statistical test. The significance level is 5%. Assays that create descriptive results will be analyzed by descriptive statistics. (AU)
