Clinical, metabolic, blood gas features and oxidative stress markers in pregnancy and postpartum of sheep: comparison between single and twin pregnancy

Abstract

The pregnancy toxemia (PT) is a disease associated with the energy deficit that affects sheep and goats at the end of gestation, usually with multiple or large fetuses. The objectives of this study are: to study the biochemical profile, blood gas, metabolic and oxidative stress of sheep during pregnancy and in the immediate peripartum (until 48 hours postpartum) and correlate them with the incidence of PT, comparing single with twin pregnancy. 60 ewes will be used, reared in semi-intensive management, healthy, of Dorper breed, with two to five years old, with an average weight of 60kg. Two experimental groups will be formed: Group I: 30 ewes, with ultrasound diagnosis of single pregnancy; Group II: 30 ewes, with ultrasound diagnosis of twin pregnancy. The experimental moments are defined as: MM - immediately after natural mating or artificial insemination (control); MG30 - 30 days of gestation; MG90 - 90 days of gestation; MG120 - 120 days of gestation; MG130 - 130 days of gestation; MG140 - 140 days of gestation; MG145 - 145 days of gestation MP - lambing day (150 days of gestation, approximately); MPP1 - 24 hours after lambing; MPP2 - 48 hours after lambing. The following procedures will be performed in all the moments: physical examination for assessment of vital functions; weighing and evaluation of body condition; transabdominal ultrasonography to confirm the fetal viability; taking samples of whole blood. Will be measured in the blood: cortisol, glucagon, insulin, T3, T4, triglycerides, cholesterol, urea, creatinine, total protein, albumin, globulin, aspartate aminotransferase, gamma glutamyl transferase; alkaline phosphatase; glucose, free fatty acids, beta-hydroxybutyrate, enzymatic activities of superoxide dismutase, glutathione peroxidase and catalase; plasma concentrations of Glutathione Total, Reactive Substances Thiobarbituric Acid and lipid peroxidation; venous blood gas analysis. In urine samples will be conducted physical and chemical tests by test strips and refractometers. (AU)
