Abstract

This application aims to get financial support to continue some of our ongoing studies which were supported by FAPESP up to last January. The studies have in common the goal of establishing new clean technologies for Organic Synthesis (Fine Chemistry) and Biochemistry (Chemistry of Peptides and Proteins) based on the combination of biocatalysis and nanotechnology. The studies proposed will be done in the following sequence: (i) study on the preparation and characterization of enzymatic magnetic nanocatalysts with synthetic and hydrolytic activities (Fe3O4@silica-thermolysin and -bovine trypsin), (ii) examination of the properties of such new Fe3O4@silica-enzymes, (iii) study on the combined use of Fe3O4@silica-enzymes and the palladium magnetic nanocatalysts Fe3O4@silica-Pd as catalysts for the synthesis of dipeptides of high scientific and commercial values, (iii) study on the feasibility of applying the Fe3O4@silica-enzymes obtained for proteolysis/sequencing/identification of proteins. Therefore, methods, techniques, procedures and protocols often used in Organic Synthesis, Peptide and Protein Chemistries, Enzymology, Proteomics and Chemistry of Materials will be employed. Due to the interdisciplinary nature of the studies, we will collaborate with experts in Catalysis and Nanomaterials, Enzymology and Organic Synthesis. Graduate and undergraduate students under my supervision will be involved. (AU)
