BONE REMODELING PROCESS EVALUATION AFTER ORTHOPEDIC FORCES APPLICATION IN RATS SUBMITTED TO CIGARETTE SMOKE INHALATION.

Abstract

Smoking habit is very common and, in most of the times, it begins early during adolescence. Among all the systemic alterations it causes, damage on cellular repair process is of great importance to understanding dental-skeletic movement. However, there is no scientific evidences related to this issue, orthodontic/orthopedic treatment and celular and biomolecular evaluations. The aim of this study will be evaluate the bone remodeling process during orthopedic mecanotherapy in rats submitted to cigarette smoke inhalation. Sixty Wistar male rats will be randomly divided into four study groups with 15 animals each: control group (intact midpalatal suture and no cigarette smoke), experimental group I (intact midpalatal suture and cigarette smoke inhalation), experimental group II (maxilar orthopedic expansion and no cigarette smoke) and experimental group III (maxilar orthopedic expansion and cigarette smoke inhalation). Each group will be subdivided into 3 experimental periods of evaluation (n=5) and the animals of these subgroups will be euthanized at 3, 7 and 10 days after orthopedic treatment. Changes in gens and proteins expression of RANK, RANK-L, osteoprotegerin (OPG), osteonectin (ONC), osteocalcin (OCC), bone sialoprotein (BSP), osteopontin (OPN) and bone morphognetic protein (BMP) 2 will be evaluated. Real-Time PCR will be used to amplify the cDNA of OPG, RANK, RANK-L, OCC, ONC, BMP-2, BSP and OPN genes that will be analyzed on the Sequence Detection Software vl.3. These data will be confirmed by protein expression evaluations applying the Western Blotting methodologies. The results of Real-Time PCR of each gen and the optic density values of immunoreactives bands will be clustered according to the differents study groups and the period of healing and will be analyzed using ANOVA with Student-t pos-test. A difference will be considered statistically significant when p<0.05. (AU)
