Improving antimicrobial photodynamic therapy for infectious disease associating peptide aurein 1.2

Abstract

According to the World Health Organization (WHO), infections by resistant bacteria affect more than 2 million people, causing more than 20,000 deaths per year in the United States and Europe. Bacterial resistance has grown over the years, however, new antibiotics development rate has declined over the past three decades, with only one new breakthrough therapeutic class since the 1980s. This growing antibiotic resistance managed to search by drugs and / or therapies that are not only capable of eliminating the resistant microorganisms, but also have characteristics which hinder the development of resistance. In this context, Photodynamic Therapy (PDT) and antimicrobial peptides are highlighted, being identified as the most promising alternatives to antimicrobial drugs for the treatment of localized infections. In this study, methylene blue and curcumin will be used as photosensitizers and 1.2 aureína peptide, a peptide of 13 amino acid residues will also be tested. Both therapeutic approaches will be assessed to increase bacterial reduction, both individually and in combination. Still, new drug delivery systems will be developed and evaluated with the aim of incorporating the system in the same photosensitizer and the peptide in order to optimize the combination therapy. Thus, this project aims to develop new drugs and pharmaceutical formulations that optimize the effect of photodynamic therapy on bacterial reduction. (AU)
