Protein malnutrition in hematopoiesis: Characterization of bone marrow stroma and Calcium Participation Assessment in hematopoiesis

Abstract

Protein-energy malnutrition is a major food problems of the world. Data from our lab and other authors demonstrate that these amendments between the PEM, is the modification of hematopoietic tissue, with changes of extracellular matrix components, changes in the cell cycle of hematopoietic stem cell / progenitor hematopoietic, reducing the production of precursor cells and hematopoietic both the erythrocyte series, as in the leukocyte series, leading to anemia and leukopenia.The leucopenia can lead to frames of infection and malnourished people have increased susceptibility to infections Protein malnutrition modifies both the specific immune response to the unspecific of the individual.We have demonstrated in murine model of malnutrition, histological changes of the bone marrow with marrow hypoplasia, changes in the extracellular matrix and alterations in the cycle of hematopoietic stem cells that remain in larger numbers in the G0 phase of the cell cycle in malnourished animals. Faced with these events, our hypothesis is that, at least in our experimental model, changes in proliferation, differentiation and maturation, observed, may result from the change in the microenvironment inducing hemopoese, compromising the cell signaling processes and the physiology of cells stem / progenitor. Thus, we propose in this study was to evaluate the kinetics of the establishment of the components of the bone marrow stroma and the participation of calcium in regulating this process emphasizing: the identification of cell types, stromal establishment of assessment and the areas of "cobblestone", detecting the production of cytokines and growth factors (IL-3, IL-6, IL-7, IL-9, IL-11, SCF, GM-CSF, G-CSF) in supernatant of cultures, evaluation of cell-cell and cell-matrix relations and extracellular (MEC) and assessment of the involvement of intracellular calcium, the stromal cells and hematopoietic stem cell.
