Antifungal activity and cytotoxicity of biomaterials, in co-culture, coated with chitosan nanoparticles containing encapsulated salivary proteins molecules (histatin 1, histatin 5 or DR9RR14)

Abstract

For this, 4 biomaterials will be assessed: 1 zirconia, 1 alloy of titanium, aluminum and vanadium (Ti-6Al-4V), polymethylmethacrylate 1 and 1 silicone addition. Specimens (14 mm diameter x 1.2 mm thick) of each biomaterial will be prepared and will be divided into two groups (n = 3): GC: uncoated (control); GR: coated. For each experimental condition are evaluated cell proliferation by Alamar Blue assay, cell viability using ATP level (adenosine triphosphate) and integrity of the cell membrane of viable cells in epithelial cells when co-cultured with species Candida. The biofilm-forming ability of the coated samples will also be assessed by the XTT assay and counting of colony forming units. Moreover, to confirm the presence of biofilm on the samples and to evaluate its thickness and presence of viable microorganisms, samples of all groups will be evaluated in confocal microscopy (CLSM). The results of cellular metabolism of viable cells after contact with the extracts of the materials studied, and the results for analysis of biofilm formation capacity, will be submitted to appropriate statistical method and the 5% significance level will be selected (± = 0.05). Moreover, for each cell viability test, the results will be compared with the negative control and treatments in different experimental groups will be classified according to the cytotoxic effect of scores 0 to 3.
