Chronic stress and adrenergic signaling in the progression of periapicopatia: a potential therapeutic target?

Abstract

This study will evaluate the importance of chronic stress (EC) in stimulation of the sympathetic nervous system and explore the pathogenic mechanisms underlying periapical lesions. Animal model of chronic stress and periapical lesion as well as cultured human fibroblasts will be used. The expression of tyrosine hydroxylase, the ±1-adrenergic (±1-AR) and ²2-adrenergic (²2-AR) receptors will be evaluated. In addition, the human periodontal ligament fibroblasts will be stimulated with lipopolysaccharide (LPS) to mimic the process of inflammation. The viability and the expression of ±1-AR and ²2-AR will be assessed. Inflammatory cytokines: Interleukin (IL) IL-1², IL-6, IL-10 and IL17 will be detected after pre-treatment with the adrenergic receptors blockers: phentolamine / propranolol, both in vitro and in vivo. The data will be evaluated by descriptive and inferential statistical tests, according to data obtained, according to their distribution, with parametric or non-parametric tests. (AU)
