Development of new bioactive restorative composites associated to nanoespheres of chitosan loaded with calcium phosphate

Abstract

One of the main reasons to replace composite resin restoration is the development of secondary caries. Aiming minimizes this problem, the development of anti-caries materials have been proposed for addition of calcium phosphates to the materials. These materials are able to release phosphates and calcium ions when exposed to low pH. Despite of to be effective in remineralization of dental structures, these materials still lack of adequate mechanical strength and more studies are needed to improve them. Based in this, the objective of this study is to develop a bioactive restorative composite with two action mode: remineralizing action and antimicrobial action. For this, It is intended: to develop chitosan nanospheres loaded with calcium phosphate; to characterize the nanospheres for light scattering, scanning electronic microscopy and thermogravimetry; to incorporate the nanospheres in photoactivated composites based on BisGMA, TEGDMA and barium glass; to characterize the developed bioactive composites regarding mechanical properties, degree of conversion, rate of calcium and phosphate ions release, chitosan release, cytotoxicity and genotoxicity. So, this project are going to develop a restorative composite, totally innovative, with potential to increase the longevity of composite resin restorations because in function of the morphology and composition of the nanoparticles, they will be able: to promote the remineralization of dental structures, due to the calcium and phosphate ions release; to present antibacterial properties due the chitosan presence; to show adequate mechanical properties and to be rechargeable with calcium and phosphates ions, releasing continuously for the oral environment.
