Considering the development of regional metastasis in OSCC is the main prognostic factor in OSCC, we aim to evaluate the role of ADAM17-overexpressing vesicles in pre-metastatic niche preparation. For that, ADAM17-overexpressing EV from HEK293 Flp-In TREX cells will be isolated, evaluated by Nano particle tracking, Cryo electron microscopy and Mass spectrometry. Male BALB/c nude mice will receive in the pad of the left anterior member 5 injections of label-vesicles originated from HEK293 following by the injection of the SCC-9 cells and/or less aggressive SCC-15 or SCC-25. The lymph nodes will be evaluated by immunofluorescence and histopathological examination. The proteome of vesicles will be examined and the effect of EV on recipient lymph nodes tissue evaluated by Target RNA sequencing. We expect that this study will provide candidate proteins that are potential hubs in oral cancer metastasis. (AU)
