Lipidomic analysis of cancer patients blood in the search for possible circulatory biomarkers

Abstract

During the development of carcinogenic cells, reprogramming of their metabolic pathways and other alterations occur due to cell growth and proliferation, energy sources, inflammation, tumor-related enzymatic metabolism and others. Such alterations are intimately related to lipid metabolism so that lipids become an excellent source of information on cell metabolism in tumorigenesis. Possible biomarkers of the response to those alterations can be obtained by means of a lipidomic strategy based on analytic tools that identify and quantify plasmatic and cellular lipids precisely, and on a large scale. This project proposes a broad and systematic (qualitative and quantitative) assessment of lipids in blood samples (plasma and erythrocytes) drawn from healthy volunteers and from patients with (colorectal and mammary) cancer undergoing treatment at the São Francisco na Providência de Deus University Hospital (Bragança Paulista, São Paulo). Two other research centers participate in the project. Chromatographic and high resolution mass spectrometry analyses will be used with an analytical platform capable of analyzing around 3,000 lipids. The differentiated molecules will be indicated by means of multivariate statistics tools. The lipidomic approach will be of fundamental importance in the discovery of biomarkers and in enabling an understanding of the molecular basis of specific pathologies. (AU)
