Search xanthine oxidase inhibitors in plant samples using different approaches of the biochromatography

Abstract

The affinity chromatography is nowadays a powerful tool for enzymatic recognition of ligands providing the establishment of highly selective, rapid and effective methods for application in highly complex matrices, such as the plant matirx. This project aims to apply two approaches of biochromatography to search for potential XO inhibitors from medicinal plants to the study of the kinetic parameters and inhibition: one using the two-dimensional chromatography with a capillary enzymatic reactor (ICER-XO) and the other with magnetic particles for "fishing" binders in plant extracts. The XO enzyme was selected for this study because it is well characterized as a target for the treatment of various disease states involving inflammation, atherosclerosis, hypertension, carcinogenesis, amongst others and also to be an urgent need for the development of more efficient inhibitors, and less toxic than allopurinol, the main therapeutic option available. The great potential of plants as new therapeutic agents, the effectiveness of these synthetic compounds study models and the involvement of CPQBA in the study of medicinal plants motivated this research project to search for XO inhibitors of natural origin.
