Abstract

Endometriosis is a complex disease that affects about 10% of women in reproductive age, causing a great economic and social impact and impairing the quality of life of these patients. Symptoms include pelvic pain and infertility. Currently, there are two approaches when endometriosis in diagnosed: medical and surgical treatment. The medical treatment includes the use of estrogen regulators and anti-inflammatory drugs that treat only the symptoms. The surgery despite being effective in removing a large part of the damage is costly and not always available to the entire population. There is thus a need for new therapies that act not only in reducing pain, but also in reducing the lesion. As Brazilian Federal Government has been currently supporting research and development studies with the Brazilian biodiversity, we intend with this research project to develop a patentable formulation of Uncaria guianensis extract and rutin. We hypothesize that these actives act on key biological processes involved in the progression and establishment of endometriotic lesions. Thus, using in vitro models, we will challenge the cells isolated from patients with endometriosis with the actives in order to evaluate their antioxidant, anti-proliferative and in reduction of cell viability effects. The best combination of the extract and rutin will then be evaluated for their effect in reducing the lesion, inflammation, cell proliferation, oxidative stress and angiogenesis in an animal (rodent) model of induced endometriosis. The chemical characterization of U. guianensis extract will be held so that we can identified the active molecule responsible for the action. We expect with this study to develop a drug that may subsequently be used in the treatment of endometriosis. (AU)
