Abstract

In this program, our main goals are: (Aim 1) To identify and validate microRNA (miRNA) expression in plasma and tumor tissue from patients, (Aim 2) To validate expression of predicted miRNA target genes in same samples, (Aim 3) To characterize metabolic changes in patient serum and sputum and integrate results with miRNA and target gene profiles. In order to achieve these goals, our research plan is to initially identify and validate biomarkers useful for early disease detection. In a second phase of our work, we will apply our scientific findings of miRNAs and metabolic changes to the characterization and validation of biomarkers with potential therapeutic application: 1) We will profile global miRNA expression in tumor and plasma samples from the same patients using the GeneChip® miRNA 4.0 Array (Affymetrix); 2) Data will be analyzed and interpreted in order to identify significantly deregulated miRNAs in tumor and plasma from patients; 3) Serum and sputum samples will be profile for metabolomic changes; 4) miRNA and metabolomic changes will be correlated; 5) Advanced bioinformatics and machine learning techniques will be applied in order to interpret changes in relation to regulatory pathways, disease development and progression; 6) Relevant clinical data will be analyzed in relation to identified changes.
