Evaluation of the effects of Polybia venom antigen 5 (Hymenoptera, Vespidae) in the recombinant form on the production of inflammatory and immunological mediators in vitro for the development of a diagnostic kit

Abstract

Although anaphylactic shock associated with immediate hypersensitivity reactions, caused by allergenic proteins present in venoms, affects a small part of the world population, Hymenoptera stings are among the main causes of systemic allergic responses, with risk of anaphylactic shock. The order of Hymenoptera insects is very broad covering wasps, bees and ants. The immune response to these insects depends on the amount of venom and the sensitivity of the individual. In Brazil there are many species of social wasps, which constitute a potential risk to humans. The social wasp Polybia paulista is very aggressive and abundant in the States of São Paulo and South of Minas Gerais, being responsible for many accidents of medical importance. Interest in the molecular characterization of these allergens has grown in the scientific world due to the promising results obtained in immunological and clinical studies in the therapeutics and prophylaxis of hypersensitivity responses. Several assays may enable a better understanding of the allergic processes and may thus improve the diagnostics by allowing the identification of epitopes and molecular regions interacting with antibodies. The present project will evaluate the potential of the p ryPoly 5 (corresponding to Poly p 5 allergen from P.paulista (Hymenoptera, Vespidae) venom and recombinantly expressed in the yeast Pichia pastoris) in the diagnosis of immediate hypersensitivity response as well as the the immunomodulatory effects on the activity of three cell types: macrophage culture Recently extracted and two cell lines of tumor origin (J744 and RAW 264.7), also derived from macrophagic lineage. Cellular and markers profile, cytokine levels )IL-1, IL-6, IL-10, IL-12, TNF-gamma and TGF- beta), evaluation of the expression of NF-ºB transcription factors in primary cultures of mouse macrophages and evaluation of cytokine mRNA expression by qRT-PCR in primary macrophage cultures of stimulated mice.
