Mitochondrial dysfunctions and DNA damage in oocytes: implications to fertility

Abstract

This proposal refers to the visit of Dr. Anand Kumas Pandey to our research group at Laboratory of Genetics and Biotechnology, Department of Genetics and Evolution, Federal University of São Carlos, during a period of 12 months. Dr. Pandey owns a large experience with the study of folliculogenesis and the pharmacologic control of it in mammals. The main goal of his visit is to strength our collaboration and implement in our group several approaches used by the researcher, mainly those involving the study of follicular development. Participation of the researcher in the projects referred below has the potential, therefore, to contribute with their development, mainly regarding the study of ovarian reserve and oocyte competence. Dr. Pandey will collaborate with Subproject 3 of our Thematic Project (FAPESP 2017/04372-0) "Characterization of the role of Rad51 in the female germline and its implications to fertility". Specifically, this researcher will contribute with analysis of the effect of Rad51 conditional knockout in the oocyte on follicular development in mice. Dr. Pandey will also be involved with two other studies. One of them refers to the study of fertility in old and young cows. This project is intrinsically related to our Thematic Project, being a complementation of it as we seek to study mitochondrial function and double-strand DNA repair mechanisms in the oocytes of these cows. The other study is related to a Regular Project (FAPESP 2017/05899-2) "Effect of knockout of mitofusins in the mouse oocyte - implications to fertility and inheritance of mitochondria". This work, which is an extension of our former project (FAPESP 2012/50231-6).In addition, Dr. Pandey will give two courses for post-graduate students. One of them will be delivered at the Programa de Pós-Graduação em Genética Evolutiva e Biologia Molecular (PPGGEv/UFSCar), while the other will at the Programa de Pós-Graduação em Anatomia dos Animais Domésticos e Silvestres (POSANATO/USP). These courses, entitled "Folliculogenesis and early development in mammals", will encompass the following topics: 1) folliculogenesis and follicular dynamics; 2) estrous cycle and methods of heat detection; 3) antiluteolytic strategies to enhance conception rate; 4) neuro-endocrinology of reproduction; 5) role of stress in reproduction; 6) maternal recognition of pregnancy; 7) fertilization; 8) placenta; 9) pregnancy diagnosis; 10) stem cell therapy. It is considered in this proposal participation of the researcher in, at least, one thesis defense and delivery of seminars in the groups led by Dr. Flávio V. Meirelles (FZEA/USP), Dr. Felipe Perecin (FZEA/USP), Dr. Juliano C. da Silveira (FZEA/USP), Dr. Pietro S. Baruselli (FMVZ/USP), Dr. Guilherme Pugliesi (FMVZ/USP), Dr. José Buratini Júnior (IB/UNESP-Botucatu), Dra. Lindsay U. Gimenes (FCAV/UNESP-Jaboticabal) and Dr. Marcelo M. Seneda (UEL). We will stimulate the researcher to be involved in several thesis defenses and collaboration with the above research groups. All activities involving Dr. Pandey will be extensively announced in our community to enhance the impact of them in the research developed in the State of São Paulo. (AU)
