Abstract

The aim of this study is to synthesize two new carrier nanosystems of antifungal drugs (DRU) (miconazole and fluconazole) based on magnetic iron oxide nanoparticles (MNPs) and chitosan (CTS), as well as to evaluate the antimicrobial effect of these nanosystems against Candida albicans and Candida glabrata. The nanosystems will be prepared by loading of DRU on MNPs coated with CTS and characterized by X-ray diffraction, infrared spectroscopy, thermogravimetric analysis and transmission electron microscopy. The minimum inhibitory concentration of the DRU-CTS-MNP nanosystems capable of inhibiting the strains in planktonic state will be determined according to the microdilution method. After, biofilms of the abovementioned species will be formed (for 24 hours) in 96-well plates and treated twice a day, during 2 days, with different concentrations of the nanosystems. Antibiofilm effect of the nanosystems will be evaluated by counting the number of cultivable cells, total biomass (crystal violet staining assay), metabolic activity (XTT reduction assay), composition of the extracellular matrix (proteins and carbohydrates) and analysis of biofilms' structure. Appropriate controls will be included in all analyzes. Data will be analyzed to employ the most appropriate statistical test. P < .05 will be considered statistically significant. (AU)
