Abstract

The heterogeneity, flexibility and cellular plasticity support the hypothesis of the association of bioactive biomaterials, with stem cells and / or bioactive molecules, as an alternative in the reconstruction of tissue losses. It is a relevant public health problem, since grafts and transplants show clinical results that are not always satisfactory. On the one hand, the lack of organs and tissues for this purpose, and on the other the plasticity of the placenta, suggests to consider the organ as an alternative source for bioactive acellularized biomaterial for the generation of new tissues. The therapeutic potential of the placenta has received attention because of its immunosuppressive and angiogenic capacity and because it is a temporary structure, mostly identical to the fetus, with a rich extracellular matrix, where hormones, cytokines, chemokines, collagens, enzymes, antioxidants and growth factors such as HGF, EGF and TGF-B. The biological responses of the placental matrix depend on the methods used for its decellularization, these affect the properties of the matrix and interfere in the choice of the cell types to be used in its recellularization. One of the main aspects of comparative placentation, ie, the establishment of the invasive (hemocor- nal), intermediate (endotheliocory) or noninvasive (epitheliocory) nature of the maternal-fetal barrier in several eutherian mammals, is essentially modulated by conditions of the extracellular matrix. Comparative studies of placenta in animal models are essential for understanding the establishment of placenta and related problems in humans. The study of the main mechanisms and differences in relation to the role of the extracellular matrix in the placental invasion of animal species will contribute to identify the factors involved in disorders during pregnancy, such as preeclampsia. Objectives: 1) to analyze the functional, structural and ultra-structural morph characteristics of the placental matrix of distinct species, more invasive placenta models - rodents, rabbits and noninvasive models such as cattle and horses; 2) choose a model of "biological scaffold" in which adult and embryonic cells can be associated to produce new tissues, including vascularized ones; 3) to analyze extracellular matrix changes important for the understanding of diseases, such as tumors and others that lead to tissue fibrosis. Extracellular matrices of bone and lung tissue will be characterized under normal conditions and with osteosarcomas and inflammatory airway disease in animal models. In parallel, changes in the placental matrix of pregnancies, produced by cloning, will be investigated, to study transcriptome and proteomics, genes and superexpressos proteins related to the increase of matrix development.
