Participation of extracellular vesicles of Sporothrix brasiliensis yeast in the immune response against sporothicosis: ex vivo and in vivo studies

Abstract

Sporotrichosis is an emerging mycosis that affects the skin, lymphatic system, and other organs of man and animals. The fungal infection has global distribution and is endemic in some regions of Brazil. The disease is caused by a complex of at least four pathogenic species, including Sporothrix brasiliensis. Some fungi are able to release extracellular vesicles (VEs) to transport molecules, which can interact with the host cell and modulate the immune response. The objective of this work is to analyze the participation of yeasts of S. brasiliensis in the immune response of dendritic cells (DCs) and in the murine model. VEs will be obtained by differential centrifugation from the supernatant of yeast cultures of S. brasiliensis. DCs derived from bone marrow will be stimulated by VEs and then cultured with yeast. Subsequently, the index of phagocytosis, and the fungal load, will be analyzed. In addition, VEs will be inoculated in Balb / c mice prior to subcutaneous infection with the fungus, and the mean diameter and fungal load of the lesion will be analyzed after different times. In this way, we hope to clarify possible virulence factors of the fungus, as well as the modulation of the immune response in vitro and in vivo, using Sporotrhix brasiliensis VEs. (AU)
