Fluoride influence on acidogenicity, polysaccharide concentration and genotypic diversity of mutans group streptococci in dental biofilm detected by AP-PCR: an in-situ study

Abstract

Although the antimicrobial effect of fluoride (F) is established, there is no evidence of a possible action of this element on the genotypic diversity of Streptococcus mutans. Thus, the objective of the present study is to evaluate the influence of F on the acidogenicity, polysaccharide concentration and genotypic diversity of S. mutans in dental biofilm formed in situ. For this, an in situ crossover study will be conducted and 16 adult volunteers will wear a palatal appliance containing human enamel blocks, subjected to 4 treatment groups: (1) Non-fluoridated dentifrice, (2) Fluoridated dentifrice with 1100 ppm F as NaF, (3) Topic application of acidulated phosphate fluoride + non-fluoridated dentifrice, (4) Topic application of acidulated phosphate fluoride + fluoridated dentifrice. The application of topic fluoride will be carried at the first day of each experimental phase. The volunteers will brush the devices 3x/day with the assigned dentifrice for each phase and drip distilled deionized water and a 20% sucrose solution 8x/day to simulate a cariogenic challenge. At the 7th day, the acidogenicity of biofilm will be determined and at the 8th day, the biofilm formed will be collected for the microbiological and biochemical analysis. For the microbiological analysis, the samples of dental biofilm and saliva of the volunteers will be inoculated in MSB. Eight typical colonies of mutans streptococci of each dental biofilm and saliva sample will be isolated and then the extraction of DNA of these microorganisms will be performed. A PCR reaction with specific primers for identification of S. mutans will be done. Then, the AP-PCR with arbitrary primer OPA-02 will be carried through and the resultant products of both amplifications will be analyzed in 1.5% agarose gel. To determine the relative mobility of each band, the photographic documentation of gels will be realized in a system of digital image and analyzed by a computer program. The biochemical analysis will include the determination of concentration of extra and intracellular polysaccharide. The representative genotypes found in saliva and dental biofilm formed under the treatment described above will be evaluated with regard to its cellular viability at pH 5.0 and 2.8; F-ATPase activity and acid production.
