Interaction between polymorphisms of genes involved in energy homeostasis and sensitivity to insulin in response to a dietary intervention to reduce body weight in obese subjects

Genetic and environmental factors are involved in the pathogenesis of obesity and type 2 diabetes mellitus (DM2). Previous studies have shown associations of our group of variants of genes of leptin, PPARgama and others at risk of obesity, DM2, dyslipidemia and coronary artery disease. Some studies indicate that genetic factors may modify the rate of reduction of body weight in response to energy restriction or weight gain for a long time. To study the interactions between polymorphisms of genes involved in energy homeostasis and insulin sensitivity and response to a dietary intervention to reduce body weight, 200 subjects will be selected obese. Individuals will be instructed to follow a reduced calorie diet for nine weeks. Before and after this period the food consumption will be evaluated by applying the Frequency Questionnaire for Food Consumption (QFCA). Will be determined anthropometric measurements and body composition by bioimpdância and will be collected blood samples for determination of laboratory parameters (glucose, glycated hemoglobin, insulin, HOMA-S, leptin, soluble receptor for leptin, adiponectin, lipid profile and inflammatory markers). It also will consider the polymorphisms of genes LEP (G-2458A and A19G), leprosy (Lys109Arg and Gln223Arg), PPARg (Pro12Ala and C161T), ADIPOQ (45T> G and 276G> T) and IL6 (-174G> C). With the data we intend to study the association of genetic polymorphisms with the biochemical variables, inflammatory, body composition and anthropometric. It also will be evaluated the possible relationship between biochemical parameters, inflammatory and anthropometric measures in response to dietary intervention. The results of this study will contribute to the understanding of gene-diet interactions in obese individuals.
