Abstract

The reduction of specific pathogens associated with caries through agents with extracts or compounds isolated from medicinal plants may be an alternative treatment for control of cariogenic biofilm. Due to its bactericidal action, evidence suggests that the oil of Melaleuca alternifolia (TTO) may be helpful in maintaining oral hygiene and the prevention of dental diseases. Objective: To investigate the effect of M. alternifolia in biofilms of cariogenic bacteria, S. mutans and L. acidophilus. Material and Methods: The first phase of this work is determination of minimum inhibitory Tconcentration (MIC) and minimum bactericidal concentration (MBC) of TTO for S. mutans and L. acidophilus. The antimicrobial effect of TTO will be analyzed on SSB (monoespécie) and DSB (multispecies) the next steps. The second phase will examine various concentrations of TTO on biofilm SSB and DSB formed on the surfaces of wells cell culture plate by the MTT technique and number of CFU / ml. In the third phase, different contact times (15s, 30, 60s) of TTO on biofilm SSB and DSB in enamel blocks will be analyzed. The gold standard, we will use 0.12% chlorhexidine repeating the same procedure. For analysis of the third phase will use number of CFU / ml, confocal laser scanning (MCVL) to observe the cells viable and nonviable biofilm and scanning electron microscopy (SEM) for the exploration of the viability of the cells biofilm.
