Abstract

Genomic imprinting, epigenetic process in which genes are expressed based on their parental origin, is involved in placental development. Many important genes for fetal and placental development are mapped on 11p15.5 region. Two imprinting control region (ICR1 and 2), including the KvDMR1 and H19DMR, regulated most of these genes. The main objectives of our project are to verify the influence of gestational age on placental epigenetic profile and the presence of mosaicism. Placentas of different gestational ages (five from first trimester, 15 from second, and 15 from third) will be selected. Additionally, four pieces from different regions of four full term placentas will be analyzed. After DNA extraction, Molecular Biology methodologies will be performed (MS-PCR, COBRA and real-time PCR approach associated to enzymatic restriction with a methylation-sensitive enzyme) for the study of the methylation pattern of KvDMR and H19DMR. The results may allow the knowledge about variations of DNA methylation during pregnancy, in special in early stages. The mosaicism research may generate data to improve future sample collections and to verify the occurrence of mistakes in the analysis of a single fragment.
