Abstract

NMR technique is one of the most powerful tools for structural elucidation special for new compounds. By other hands, the classic phytochemistry protocol for compounds isolation and identification are very establish in the literature. However this protocol have been discuss specially with concern about time consume, money spent and specially the environment impact caused by the use of a large quantities of chemical solvent. On alternative proposed in the last years is to use of hyphenation techniques. In this project the first objective is to use, for the first time in Brazil, LC-NMR and LC-SPE-NMR hyphenation techniques to study the metanolic extract of Spilanthes acmella to characterize the most important compounds presents in it and using a minimum extract manipulation. Whit this approach it will be possible to compare the classical methodology with the hyphenation one special focus on the time consume, number of characterized compounds and the money spent with solvent, time machine, etc. in all the study. The second objective is to have the metabolic profiling for 27 Spilanthes acmella genotype sample using 1H NMR and chemometrics techniques, and compare with the HPLC study done by other partner group which are involved to increase the amount of spilanthol. The choice for plant target Spilanthes acmella is because its commercial interest specially the amount of spilanthol present in this genera which is using in the pharmaceutical and cosmetic industry.
