Abstract

The cachexia syndrome is characterized by several homeostatic perturbations including depletion of muscle and fat tissue, hypoglycemia, accompanied by progressive wasting, weakness and anorexia. The cancer cachexia syndrome, which affects a large proportion of patients with solid tumors, is associated not only with decreasing quality of life but also with a shorter survival time and poor response therapy. The imbalance in glycemic homeostasis and the increase of inflammatory response caused by tumoral development are among the main factors involved in the establishment of the cachetic syndrome. However, there are no studies that evaluate changes in the endocrine pancreas that may interfere with the imbalance of glycemic control in individuals with cachexia. In previous studies we noted the establishment of cachexia in solid Ehrlich carcinoma bearing-mice. These animals have reduced insulin secretion and decrease in expression of key proteins in insulin signaling pathway, like PKA-±, M3 e AKT. With all these results, numerous opportunities for study appeared, among them the establishment of inflammatory response in islets. Thus, the purpose of this study is to evaluate components of islet inflammatory response of cachetic solid Ehrlich carcinoma bearing-mice. For this, male adult Swiss mice will be subcutaneous inoculated with solid Ehrlich tumor and sacrificed 14 after tumor implantation whilst control group will receive saline alone (CTL). Real time PCR will be performed in islets to analyze for evaluate gene expression of toll-like receptors 2, 3, 4, and 9, Western blotting for determination of pro-inflammatory cytokines expression such as TNF-±,IL-², IFN-³, IFN-±, IL-6 and IL-8, and immunochemistry for detection of macrophage infiltration. The results will be expressed like Media±SE. Significance of the differences will be evaluated by analysis of variance followed by Tukey's test. The significance level will be P<0.05.
