Influence of co-administration of fluoride from drinking water on the metabolism and uptake of lead in rats: dose-response study

Abstract

The aim of this study is to evaluate the metabolism and uptake of Pb in rats sco-administered with F from drinking water since the beginning of gestation, taking into account different doses of F and Pb as well as two types of fluoridated salts (H2SiF6 or NaF). For this purpose, 320 Wistar rats will be divided into 32 groups (10 animals each). The differences among the groups will be the doses of Pb (0, 1, 10 or 30 mg/L) and of F (0, 5, 50 or 100 mg/L, each as NaF or H2SiF6) in the drinking water to which females rats and their offspring will be co-exposed from 1 week prior to mating until offspring is 80 days old. After the experimental period, blood and femurs will be collected for F (electrode, after HMDS-facilitated diffusion) and Pb analysis (ICP-MS). Samples of urine and feces (24 hours) of the animals housed in metabolic cages (n=6 per group) will be collected every 14 days since weaning until euthanasia. These will also be analyzed for F (electrode, direct method for urine and after HMDS-facilitated diffusion for feces) and Pb (ICP-MS) in order to determine absorption and excretion of F and Pb in the different groups. Data will be subjected to appropriate statistical tests, after checked for normality and homogeneity (p<0.05). (AU)
