Abstract

The most effective control of pain has motivated the search for new local anesthetic formulations. The association with carriers has been proposed in order to prolong the anesthetic effect and reducing its toxicity. New anesthetic formulations produced a low systemic toxicity and longer duration of anesthesia. Adverse effects were also observed as the appearance of inflammatory painless without apparent cause. This inflammatory response without unknown nature could be triggered by the release of proinflammatory cytokines. The application of these formulations in humans requires biocompatibility and no important adverse effects. Thus, the objective of the study is to observe the effects of local anesthetics and associated with free carriers on the modulation of the inflammatory mediators IL-6, IL1-±, IL-8, TNF-±, IL-10 and PGE2, in addition to its action on cell viability in cultured oral epithelial cells and human gingival fibroblasts. The cells will be exposed to different concentrations of anesthetics bupivacaine, lidocaine, ropivacaine, and benzocaine free or associated with liposomes or cyclodextrin. Quantification of cytokines will be done using the capture ELISA after 6h and 24h of exposure to anesthetics. Cell viability will be estimated after the exposure periods and compared to the untreated control group. Statistical analysis will be performed by ANOVA (Turkey test) or Kruskal-Wallis test (Dunn) depending on the distribution of data and homoscedasticity of variances. The significance level is 5% and the statistical package will be BioEstat 5.0. (AU)
