Role of LH during induced luteolysis and characterization of luteal sensitivity to PGF2alpha during estrus cycle and treatment with IFNt

Abstract

The PhD project aims to enhance studies on luteal regression in cattle. For this three researches will be conducted. Experiment 1: aim to evaluate LH effects on the rescue of P4 secretion after challenge with cloprostenol sodium at D7 of estrous cycle (D0=estrous). Caracu cows on D7 will bee submitted to the following treatments: Group 1 (G1): saline, G2: saline associate to GnRH antagonist (acyline), G3: 1/6 of cloprostenol conventional dose, G4: 1/6 of cloprostenol conventional dose after acycline administration, G5: cloprostenol conventional dose. From 12 to 20 hours after treatment blood samples will be collected every 1 hour to LH assays. Experiment 2: aim to evaluate CL sensibility to PGF2 subdoses at initial (D7) and medium (D12) phases. Caracu cows will be placed in the following groups: G1: saline D7, G2: saline D12, G3: 1/6 of cloprostenol conventional at D7, G4: 1/6 of cloprostenol conventional at D12. Experiment 3: aim to evaluate CL sensibility after intra uterine exposure of bovine Interferon Tau (IFNÄ). Caracu cows will be placed in two groups: G1: intra uterine infusions of IFNÄ from D9 to D12 following by PGF2± challenge at D12. G2 (control group): PGF2± challenge at D12. In all experiments immediately after treatments (0hour) and after this every 8 hours until 48hours (M48) the animals will be submitted to transrectal ultrassonographic evaluation at mod B (luteal volume) and Power Doppler (luteal blood perfusion). Blood samples will be collected for P4 assays. (AU)
