Abstract

Bacillus subtilis spores have a record of use as probiotics and, presently, represent an alternative vaccine delivery approach. In additions, recent studies have demonstrated that B. subtilis spores are endowed with strong adjuvant effects for vaccines based on purified proteins. The aim of the present project is to evaluate the role of B. subtilis spores as adjuvants applied to DNA vaccines delivered via different administration routes, with emphasis to the intradermal (i.d.) route. As an experimental model we employed a DNA vaccine (pgDE7) encoding the E7 protein of the type 16 human papilloma virus (HPV-16) genetically fused to the type 1 human herpes virus (HSV-1) gD protein. The spore adjuvant effects will be monitored by the induction of antigen-specific immune responses and by the anti-tumor protection effects in C57BL/6 mice previously transplanted with transformed cells (TC1 cells) carrying oncogenes encoding the HPV-16 E6 and E7 proteins. Our results have indicated that bacterial spores exert strong adjuvant effects on the immune responses induced by DNA vaccines and may represent, therefore, an useful alternative for increasing the performance of this kind of vaccine.
