Evaluation of potential chemopreventive oil pequi (Caryocar brasiliense Camb.) by medium-term test

Abstract

Medicinal plants and the research of active substances in plants is a field that has been extensively approached recently, especially for researchers looking for protective effects in human and animal carcinogenesis. The Brazilian native flora has several plants that have a great research potential in this subject, one of these being the Pequi (Caryocar brasiliense Camb.). This fruit from the central region of Brazil contains several antioxidant substances in its meat, mainly in its oil extract, which are related with the reduction of the risk of degenerative diseases. In this project we propose to evaluate the chemopreventive potential of pequi oil against preneoplastic hepatic lesions chemically induced by diethylnitrosamine in the infant mouse model in a mid-term test. Diethylnitrosamine (DEN) will be intraperitonially injected in 15 days-old mice (10µg/g) as initiator. Five experimental groups will be formed (GI= positive control, DEN); GII, GIII and GIV (DEN + pequi oil by daily gavage, 100 mg, 400 mg, 1000 mg respectively after the 30°day to the 119° day) and GV (control, pequi oil 400mg). The evaluation of the effectivity of the pequi oil in the protection against the development of preneoplastic lesions will be done by the histopathologic analysis of the sections and quantification of the altered hepatocites foci (AF) among group, aided by the analysis of the immunohistochemical expression of cytokeratins 8/18 (CK 8/18) e CK 19.
