Potential cytotoxic of national denture base acrylic resins: influence of heat treatment and water storage

Abstract

Many studies have been made to verify the acrylic resins' cytotoxicity. However, those studies are based on imported resins, not widely used in prostheses laboratories in Brazil. Thus, the objective of this study is to evaluate the effectiveness of heat treatment and additional water storage in reducing national denture base acrylic resins cytotoxicity, through the MTT assay, which evaluated mitochondrial activity in cell proliferation. Nine disc-shaped specimens (10 x 1 mm) of each denture base resin (Lucitone 550, Clássico, Clássico Onda Cryl, Vipi Cyril, and Vipi Wave)will be prepared according to the manufacturers' recommendations and stored in distilled water at 37ºC for 24 or 48 h. After polymerization, after 24 or 48 hours of storage, the specimens will be assigned to 3groups: 1) post-polymerization in a microwave oven for 3 min at 500 W; 2) post-polymerization in water-bath at 55º C for 60 min, and 3) with outpost-polymerization. For the preparation of eluates, 3 discs will be placed into a sterile glass vial with 9 mL of Eagle's medium and incubated at 37ºCfor 24 h. The cytotoxic effect of the eluates will be evaluated by MTT assay.(AU)
