Abstract

The genus Koellensteinia Rchb. f. (Orchidaceae) encompasses 17 South American species. Problems related to the delimitation of its species are still present and recent phylogenetic studies denote that the genus, as presently conceived, is not monophyletic, and its relation with the nearest genera is uncertain. This project aims to: I) propose a phylogenetic hypothesis for Koellensteinia and related genera; II) to reconstruct a biogeographical history of the group as a whole and that of the K. eburnea - K. tricolor complex in particular; III) to determine the chromosome number of the studied species and reconstruct the cariotipic evolution of the group; IV) to produce a taxonomic revision of Koellensteinia based on the results achieved. The specimens will be gathered in the regions Meadle-West, North, Northeast and Sudeast of Brazil, besides of Bolivia, Colombia, Ecuador, Peru and Venezuela. The DNA of the sepecimens will be extracted from fresh or desiccated leaves. First off the regions ITS, matK, rbcL and trnL-F will be used to elucidate the position of Koellensteinia in the subtribe, because these are the regions formerly used for other genera of Zygopetalinae. A trial of other nuclear and plastidial regions will be performed, seeking for regions showing higher mutation rates for explain the infrageneric and evolutinary relationships. The phylogenetic and biogeographic analises will be performed from the aligned sequences, using bayesin, parcimony and likelihood methods. The molecular dating will use bayesian methods with the inclusion of fossiles for molecular calibration. At the same time a cytogenetic analisis will be accomplished. (AU)
