Abstract

The present study aim to perform a gestational assessment in domestic cats previously treated with deslorelin acetate and to produce in vitro embryos through oocytes ovaries of domestic cats under treatment with deslorelin acetate. For this two studies will be performed. Experiment 1: 5 domestic cats will be used. The animals will remain in quarantine for at least 30 days and will be accompanied to sexual behavior, vaginal cytology (VC) every 3 days and fecal progesterone concentrations every 7 days. When in interestrous/diestrous phase the contraceptive treatment with deslorelin acetate (Suprelorin®, 4.7mg/animal) will begin. Queens will be monitored through VC, sexual behavior and hormonal assays every 7 days for at least 180 days. After this period, when females present estrous VC and sexual behavior, queens will be kept together with proved fertile tom cat, to mating. The females will have they feces collected every 48 hours after this moment. Pregnancy will be confirmed after 30 days of the last mating and gestational assessment will be performed through ultrasound exames (every week) and progesterone assay (every 48hours) until parturition. In case of abortion the females will be accompanied and caesarean and OSH will be performed. In this cases the uterus and ovaries will be analyze through histopathological and imunoistochemistry (sexual hormones receptor) exams. Microbiological exams will be performed to exclude or confirm possible causes of infectious abortion. Experiment 2: 20 queens will be placed in two groups: treated (A) and control (B). Three tom cats will be used for semen collection. The quarantine period, sexual behavior observations, VC, progesterone assays and contraceptive treatment on group A (n=10) will be performed as previously described on experiment 1. However after 180 days of contraceptive treatment all females will be submitted to OSH for oocyte recovery and posterior IVM and IVF. After embryo in vitro production fresh and warmish blastocysts will be submitted to TUNEL technique with "Kit in Situ Cell Death Detection Kit Fluorescein" (Roche® - 11684 795 910, Mannheim, Germany). In group B (control) the queens (n=10) will be evaluated as described at experiment 1, but at interestous/diestrous these females will not receive contraceptive treatment and will be submitted to OSH. The same analyses described for group A will be performed. Data will be submitted to appropriated statistical test.
