Permeation studies of lidocaine across mucosal oral epithelium

Abstract

Permeation studies in vitro and ex vivo performed in Franz diffusion cells are important during pre-clinical phase of a new developed medication because it is possible to preview how this drug will permeate through the barrier, i.e. mucosal or skin, allowing pharmacokinetics and a disponibility improvement. The literature has shown that permeation studies across oral mucosa are performed using the pig esophagus mucosa model. However, this tissue has little or no keratinization being the esophagus a limited model to evaluate the performance of formulations for dentistry use. There are other regions of the mouth with different levels of keratinization (palatal mucosa and gingival) which also target sites for drugs such as local anesthetics, antimicrobial agents, corticoids, antivirus, etc. The objectives of the present study are to compare the permeation profile of lidocaine gel across different regions of pig oral mucosa epithelium (palatal, tongue, cheek, and gingival) with pig esophagus' mucosa epithelium in vertical Franz diffusion cells (fresh and frozen tissues) and to perform a histological assessment of those barriers (fresh and frozen tissues). We hope to develop methods that could be useful for permeation studies for drugs used in dentistry, to improve their pharmacokinetic and pharmacodynamic performance.(AU)
