Abstract

Parasitic infectious diseases, considered neglected diseases, represent a major public health concern in many countries and regions. The available drugs in therapy are, generally, toxic and have low efficacy. Therefore, the discovery and design of new chemotherapeutic agents are urgently needed. In this context, prodrugs and targeted drugs dendrimers may be useful. However, the active compound must be released from the dendrimers and the cleavage prediction represents a challenge for the scientific community. Considering that, the objective of this work is the creation of the Sector for Drug/Bioactive compounds Release from dendrimer compounds, prodrugs and targeted drugs, potentially active in neglected diseases at the Laboratory of Design and Synthesis of Chemotherapeutics Potentially Active in Neglected Diseases - LAPEN -Faculty of Pharmaceutical Sciences, University of São Paulo. This is an essential research area for evaluating the hydrolysis of labile linkage of the dendrimer compounds using different pH, in human plasma and rat liver homogenate. The molecules are being synthesized and they are part of LAPEN projects, such as master, doctoral and post-doctoral projects. This área is innovative and very interesting, thus, this new sector will have a great importance in developing of the group's work. In addition, molecular dynamics simulations of the complexes between esterase and derivatives, to assist in understanding the practical results, will be performed. The purpose of those studies is to aid in the understanding of the practical results and to complete the research previously developed, using molecular modeling, whose results have been recently published.
