The aim of this study is to evaluate the cytotoxicity and solubility of the mineral trioxide aggregate (MTA - Angelus, Londrina - PR, Brazil) and associations with AH Plus (Dentsply, Brazil); castor-oil bean based experimental sealer (Polifil - Araraquara Polímeros Quimicos Ltda., Araraquara - SP, Brazil) and Copaifera sp. oil-resin based experimental sealer (Cop Endo, Brazil). For the cytotoxicity test, sealers will be prepared and kept in contact with the culture medium according to the setting time (0 hour, 24 hours, 7 days, and 30 days). Chinese hamster fibroblasts (V79) will be exposed to different dilutions of the sealers extracts for 24 hours and the cell viability will be assessed by MTT test. For the solubility test, 12 specimens of each retro filling sealer will be prepared using Teflon molds. The specimens will be stored in the oven at 37°C with a relative humidity of 100% for 7 days for the completed set. The specimens will be weighted and kept in capped bottles with distilled water in the oven for 24 hours. After that, they will be dried and weighed again. The same procedure will be repeated for periods of 48 hours, 1 week, and 1 month. The results will be analyzed by the statistical tests: ANOVA and Tukey, with 5% significance (p<0,05).(AU)
