Abstract

The procedure for sample preparation is crucial for the analysis of drugs present in biological fluids in order to carry out a prior sample preparation endogenous interferences and obtain a pre enriched fraction of the analyte in question. Conventional methods for the preparation of samples such as liquid-liquid extraction and solid phase extraction , are still the most used . However , miniaturized extraction techniques have been gaining ground in the analytical area due to its numerous advantages . These employ a very small amount of sample , a minimal amount or no organic solvents ( green chemistry ), potential automation , reduced in steps of sample preparation and speed. Thus, the objective of this project is to evaluate the dispersive liquid-liquid microextraction ( DLLME ) for review of levetiracetam ( LEV ), an antiepileptic, and analysis of risperidone ( RSP ) , an antipsychotic , in human plasma by chromatographic techniques spectrometry mass . Plasma Pretreatment to remove the plasma proteins is performed by ultrafiltration using Amicon filters ( pore size of 10 kDa . Then 1 ml of clear permeate which is subjected to DLLME parameters will be assessed extractor type and volume of solvent and dispersant , pH medium , effect of ionic strength and extraction time. DLLME After parameters have been optimized , the method will be validated in accordance with the official guides , ANVISA (2012 ) and EMEA (2011 ) and applied to a plasma sample patient under treatment.
