Abstract

The renal tumor is the eighth most common cancer and highly lethal. It is a histologically heterogeneous disease, with the clear cell carcinoma (ccRCC), the most common histological subtype, comprising 70% to 80% of renal tumors. Although nephrectomy is a well established treatment, many patients develop metastasis. The ccRCC in the early stages, may change biological losing epithelial differentiation, a process called Epithelial Mesenchymal Transition (EMT). EMT is a change in molecular and cell coordinate, it is defined as a decrease in cell-cell adhesion, the areas of occlusion, the apical-basolateral polarity and mobility of the acquisition, aiding in the process of metastasis. This project aims to analyze the expression by RT-PCR (qRT-PCR) and immunohistochemical analysis of genes related to epithelial mesenchymal transition, and to evaluate the profile of genes methylation by MSP down-expression in renal cell carcinoma clear and renal carcinoma cell line treated and not treated with demethylating agent. Thus, the molecular and genetic characterizations allow a specific correlation between gene expression the methylation profile and the process of epithelial mesenchymal transition, in addition to the identification of potential markers that may contribute to the development of new therapeutic strategies.
