A. baumannii is a common pathogen associated to health care infections in Brazil, due to its capacity to survive in adverse conditions and develop resistance to broad spectrum antimicrobial agents. The high resistance rate combined to together with the absence of new antimicrobial options turned polymyxins the last line antimicrobial drugs to treat Gram negative MDR infections. The aim of this study is to analyze the transcriptomes of the polymyxin B susceptible and resistance A. baumannii clinical isolates. The strains will be submitted to the polymyxin B exposure and the susceptibility profile will be performed. The transcriptomes of the clinical isolates and their induced mutants will be determined and analyzed by comparision. The transcriptome results will be confirmed by Real Time PCR. The membrane potential will be measured before and after the polymyxin B exposure. Virulence experiments will be performed by in vivo model C. elegans.
