Abstract

The reproductive potential of bulls is important for commercial viability and success of livestock breeding. Semen analysis is the main method of diagnosis of fertility, however does not assess the internal structure of the cell and sperm chromatin. The integrity of DNA is essencial, since chromatin defects are incompatible with fertility. The aim of this study is to evaluate and compare the chromatin packing and sperm head morphometry of frozen semen samples of young, adult and senile Bos indicus Nelore bulls and the influence on fertility. A total of 40 animals kept in an Artificial Insemination Centre will be divided into three groups: Young Group (1.8 to 2 years), Adult Group (3.5 to 7 years) and Senile Group (8 to 14.3 years). The ejaculates will be frozen in accordance with established standards of the Artificial Insemination Centre. The thawed semen samples will be evaluated with toluidine blue, which analyze the chromatin packing and sperm head morphometry simultaneosly, cromomicin A3 to analyze the protamination and in vitro production to analyze the embryonic development.In order to compare the differences in semen parameters with the age groups the analysis ofvariance (ANOVA) will be used and means compared by Tukey test. Within each group the correlation of variables will be analyzed.
