Whey proteinograma with emphasis in acute phase proteins as a method of diagnosis of mastitis and for monitoring cows with mastitis under treatment

Abstract

The present study aims to evaluate whey proteinogram, including concentrations of acute phase proteins (AFP´s) as a method of diagnosis of mastitis and for monitoring cows with mastitis under treatment. The animals will be divided into two experimental groups: G1 (n = 20): mammary quarters of healthy cows (negative bacteriology, negative CMT and SCC <100,000 cells / mL) and G2 (n = 20): mammary quarters of cows with recent clinical mastitis (presence of cloths and positive bacteriology). The animals of G2 will undergo treatment soon after diagnosis of mastitis, being accompanied by physical examination (palpation of the mammary gland and the presence of clots in milk) and bacteriological examination at the following times: on the day of the finding of mastitis before first application of medicine (M1) and the first (M2), third (M3), fifth (M4), seventh (M5) and ninth (M6) days after the end of treatment. Physical examination of the mammary gland, as well as black sieve cloth test and California Mastitis Test (CMT) will be performed before sampling. After asepsis of the teats, samples of 50 ml of milk from each quarter will be collected into sterilized jars for microbiological isolation and for the analysis of protein fractionation on SDS-PAGE. Samples of 50 mL of milk from each mammary quarter will also be collected in sterile plastic vials containing preservative suitable for the somatic cell count (SCC), which will be performed in infrared electronic counter. The variables will be subjected to analysis of variance (ANOVA) and Tukey's test (p <0.05) for comparison between pairs of means at 5% level of significance. (AU)
