Metabolism in Annona emarginata (Schltdl.) H. Rainer seeds during immersion treatments in aqueous solutions

Abstract

The curve acquisition seed water is used as a reference for determining the time of immersion treatment. However, the immersion time and/or the water content achieved by the seed may change seed germination percentage. Thus, Gimenez (2012) found that immersion in aqueous solutions, with or without plant growth regulator, of seed Annona emarginata (Schltdl.) H. Rainer, species used as rootstock for commercial Annonaceae, until they reach different water contents, change the concentration of soluble sugars and germination. Thus, the goal of this project is to assess how the immersion of the seeds until they reach certain levels of water alters the mechanisms involved in the germination process of A. emarginata (Schltdl.) H. Rainer. The seeds of A. emarginata with water content of 10% will imbibed on paper moistened with water or immersion in water until they reach levels of 15, 20, 25, 30 and 35% water. In a second experiment, the seeds will be soaking in solutions with osmotic potentials of 0, -0.6 and -1.2 MPa until reaching levels of 15, 20, 25, 30 and 35% water in order to verify the effect of time in the acquisition of different water contents. During the experiments will be monitored water potential, water content, ethanol production and respiratory activity of the seeds. As the seeds reach each water contents will be held the germination test, anatomical and ultrastructural analysis of the embryo, analysis of soluble sugars (sucrose, glucose, fructose and myo-inositol), starch, proteins (albumin, globulin, glutelin and prolamin), lipid, enzyme activity (peroxidase, superoxide dismutase, catalase and ascorbate peroxidase) and plant hormones (Gibberellin and Abscisic Acid).
