Abstract

Periodontitis is a polymicrobial infectious disease that has been reported as being a global health problem by the WHO. Research on the etiology of periodontitis has lasted for more than three centuries. Advances in microbiological diagnostic techniques have shown, with increasing resolution, the huge complexity of the interaction between the oral microbiota and the host. Molecular biology techniques have allowed for the detection of not yet cultured microorganisms in oral plaque samples, which has significantly increased our understanding of the etiology of periodontal disease. However, culture-independent techniques for cataloguing microbial species have mainly been based on PCR amplification of phylogenetic markers, e.g. 16S rDNA, which has generated intensive discussion in the literature concerning limitations related to the initial amplification steps. Next generation sequencing techniques have allowed for the direct sequencing of metagenomic DNA, reducing the biases incurred from gene cloning techniques. Until now, this approach has been used in only a few studies concerning the oral microbiota and only one has evaluated subjects without (n=3) or with (n=2) periodontal disease. These studies have yielded results that describe an oral microbial diversity orders of magnitude larger than from studies employing PCR and cloning. Thus, the present study aims to evaluate in detail the composition of subgingival plaque associated with periodontitis. This will be the first study to assess individually (non-pooled) plaque samples in subjects with and without periodontal disease using a metagenomic DNA direct sequencing approach.
