Abstract

Given the importance of sperm upon fertilization and early embryonic development, know the effects of changes resulting from cryopreservation, as the production of reactive oxygen species (ROS) and their impact on this cell is critical to the improvement of biotechnologies such as in vitro fertilization (IVF). The aim of this study will be evaluate the impact of different concentrations of agents that induce oxidative stress on sperm and the effect of oxidative stress in the early stages of embryo development. First, treated and control sperm will be stain with fluorescent probes and evaluate by flow cytometry capillary. Variables related to sperm chromatin structure (acridine orange), mitochondrial potential (JC-1), membrane integrity (FITC-PI) and sperm capacitation (chlortetraticlin), and some of then related to oxidative stress (dichlorfluorescein, CellROX®, and BODIPY-C11) will be analyze at this stage. Besides the microcapillary flow cytometry, part of the sediment will be separate for conducting the test of thiobarbituric acid reactive substances (TBARS). In a second phase, groups of sperm treated with agents that induce oxidative stress and control will be use in the production of in vitro embryos, looking for verify the impact of different levels of oxidative stress on in vitro fertilization and early embryonic development. For this, the embryos will be evaluate for cleavage and the rate of formation of pronuclei.
