Osteoporosis is a osteometabolic disease mainly characterized by increased bone porosity with reduced bone mineral density. Epidemiologically significant, this pathology has shown increasingly high prevalence in the world, a fact which can be explained by a higher life expectation, based on this, osteoporosis has concerned public health entities worldwide. Current treatments can be divided into anabolic therapies, which aim to increase bone formation, or anti-resorptive therapies, which reduce bone matrix reabsorption. Studies have shown therapeutic properties in the consumption of yerba mate (Ilex paraguariensis St. Hil. var Paraguariensis), effects as hypolipidemic activity, weight reduction and anti-inflammatory activity were emphasized. More recently, improvements in bone matrix density in women post-menopause were associated with yerba mate infusion consumption, however in vitro studies in the literature describing the effects of Ilex paraguariensis in bone biology are still absent. As seen, the objective of this study is to assess the direct effect of soluble extract of Ilex paraguariensis during differentiation of mesenchymal stem cells from bone marrow stroma of young Wistar rats into osteogenic lineage. For this purpose, different concentrations of the extract of Ilex paraguariensis will be added to the culture medium containing the classical osteogenic inducer (ascorbic acid, ²-glycerophosphate and dexamethasone), which will be used for osteogenic differentiation assays where the following parameters will be assessed: cell proliferation, total protein, alkaline phosphatase and mineralization. Furthermore, TNF-alpha dosage, a proinflammatory cytokine, will be performed to evaluate the possible immunomodulatory properties of the extract.
