Abstract Due the dissemination of infections caused by MRSA, the need to create an effective vaccine against this pathogen. U.S. only clone USA300 causes 80% of CA-MRSA infections. In preliminary studies in the laboratory of the investigator Binh An Diep found that lipoproteins corresponding to 70% of the peptides found in the cell wall of USA300. Little is known about the role of the six most abundant lipoproteins found in the pathogenesis of S. aureus, and if they have a good standard for vaccine production. Thus, this study aims to determine the protective efficacy of immunization with recombinant lipoproteins in an experimental model of bacteremia. For the production of recombinant proteins will use the cloning and expression of lipoprotein using vector pTX15. 7 will groups be used of 15 A/J mice each group to be immunized with each of the six recombinant lipoproteins or placebo. Blood samples will be collected retro-orbital on days 0,11 and 20 for measuring whole antibody titer against each type of protein by ELISA. On day 22, the mice receive 5x107 CFU of wild strains USA300 and SF8300. After the mice will be monitored for seven days and euthanized, will be evaluated survival, bacteria counts, abscess formation and histopathology of the kidneys. (AU) | |
